RANKL links arterial calcification with osteolysis.

چکیده
Vascular calcification is a pathological sequence of events that has similarities to the normal physiological process of osteogenesis.1,2 It is thought to result from an imbalance in both local and systemic inhibitors and promoters,3,4 which occurs as a consequence of chronic inflammation, hyperleptinemia, and a deregulation of various bone-regulating proteins. It is common in patients with diabetes and renal failure and in the elderly.4,5 The fact that coronary artery calcification is such a major health problem and the recognition that it is highly correlative with mortality risk6 makes the understanding of vascular calcification a worthy and exciting area of study.

The RANKL/RANK/OPG Signaling Pathway Mediates Medial Arterial Calcification in Diabetic Charcot Neuroarthropathy
OBJECTIVE The receptor activator of nuclear factor-κB (RANK), RANK ligand (RANKL), and osteoprotegerin (OPG) signaling pathway (RANKL/RANK/OPG signaling) is implicated in the osteolysis associated with diabetic Charcot neuroarthropathy (CN); however, the links with medial arterial calcification (MAC) seen in people with CN are unclear. This study aimed to investigate the role of RANKL/OPG in MA...
متن کاملExenatide can inhibit calcification of human VSMCs through the NF-kappaB/RANKL signaling pathway
BACKGROUND Arterial calcification is an important pathological change of diabetic vascular complication. Osteoblastic differentiation of vascular smooth muscle cells (VSMCs) plays an important cytopathologic role in arterial calcification. The glucagon-like peptide-1 receptor agonists (GLP-1RA), a novel type of antidiabetic drugs, exert cardioprotective effects through the GLP-1 receptor (GLP-1...
متن کاملWear particles enhance autophagy through up-regulation of CD147 to promote osteoclastogenesis
Objective(s): The study aimed to uncover the underlying mechanism linking wear particles to osteoclast differentiation, and we explored the effect of titanium particles of different sizes on CD147 expression and autophagy in macrophages. Materials and Methods: Effects of titanium particles on CD147 and RANKL mRNA were detected by QPCR; protein level of CD147 and Beclin-1 were detected by Wester...
متن کاملRole of OPG/RANKL/RANK axis on the vasculature.
Vascular calcification, a degenerative process considered in the past to be a passive procedure, has now been suggested to be related to ossification. Many proteins responsible for bone formation have been identified on the arterial wall. The OPG/RANKL/RANK axis, responsible for ossification and bone mineralization, seems to play a major role in vasculature and atherosclerosis. Mice lacking OPG...
متن کاملOmentin-1 attenuates arterial calcification and bone loss in osteoprotegerin-deficient mice by inhibition of RANKL expression.
AIMS Omentin-1 (also known as intelectin-1) is a recently identified visceral adipose tissue-derived cytokine that is inversely related to obesity. Our previous study showed that omentin-1 inhibits osteoblastic differentiation of calcifying vascular smooth muscle cells (CVSMCs) in vitro. This study was undertaken to investigate the effects of omentin-1 on arterial calcification and bone metabol...
